To analyze the role of such transcription factors in controlling the decision self-renewal versus differentiation we like to functionally characterize identified candidates. In the course of the project over-expression and loss- of function studies of corresponding factors should be performed. For the over-expression studies cDNAs encoding the transcription factors of choice should be cloned into lentiviral vectors. To reduce the activity of the transcription factors appropriate RNA-interference mediating plasmids should be created.

After transducing non-induced USSC with corresponding lentiviral plasmids, effects on the cell fate of USSC should be studied in comparison to controls using established read out technologies.

After identifying any factor whose altered-expression results in a recognizable effect on the biology of USSC, it will be analyzed in detail. To identify down stream targets of such factors global gene expression analysis should be performed in comparison to appropriate controls. By performing these expression analyses at different time points post transfection, we like to identify early, intermediate and late responding genes. During the course of the project distinct transcription factors belonging to the group of early responding genes should be selected, and analyzed.
